The corporation unveiled its intentions after a number of failed attempts to sell the two properties on the east and west of the Hume Freeway earlier this year, reports the Sydney Morning Herald.
Located approximately 25 km from the city's central business district, the site is expected to garner around $10 million (£6.5 million) at auction, making it one of the highest priced properties sold in recent years.
It is situated immediately south of the Amaroo Road overpass and close to the Highlands housing estate established several years ago in Mount Ridley.
The purpose of Vicroads is to deliver economic and environmental benefits to communities across Victoria through management of the road network, along with its use as an important part of the overall transport system.
It aims to achieve reductions in the number of road crashes and the cost of incidents caused by accidents, while building relationships with customers.
